All-time...I like Eddie Van Halen or Steve Vai.
Honorable mentions to Tom Morello and Buckethead for more modern times. Maybe even Prince.

As far as rock guitar as most know it, there is Hendrix and then there's everybody else. The virtuoso guys (Vai, Malmsteen, that other guy) are egotistical wankers that play wank on wank guitars and HATE MUSIC. I hate them so much I just had to bring it up. Hard to argue with Van Halen even if I hate his/their music and, of course, Page could play pretty much anything and do it as only he could. Neil Young is a personal favorite as well... But let's not get ahead of ourselves; Hendrix built this house and he's still the king [of rock guitar]. The opening minute or so of Voodoo Child (Slight Return) puts any debate to rest * ! *

For folk, I'll take Nick Drake. Bizarre tunings, plucking patterns, and chord progessions that are fresh to this day.

Doug Martsch is up there for me, but that has a smattering of preference built in. Otherwise, I'd say Jimmy Page.

Actually, both of these guys are similar in that they're incredibly dynamic, original, and TALENTED, but both tend to be sloppy live players, if not sloppy in general.

And while they may not be virtuosos like the above, you do have to give props to Tony Iommi, Angus Young, and Jimmy Page. They are the riff-masters!

And I'm lucky enough to say I've seen 6/10 of the above in concert. And I don't know if it's the Gibson SGs, but Tony Iommi and Angus Young have the cleanest sounding guitars out of all of the above. Of course, it could also be their budgets  * smile *

Jimi Hendrix first, then Jimmy Page (no one likes Jimmy Page??!!) and then Eddie Van Halen for me. I am also a big fan of John Frusciante from Red Hot Chili Peppers.
Lastly, Kirk Hammett of "Metallica" and Slash

Honorable mentions to Tom Morello for being a virtuoso in effects driven guitar work and Yngwie for being a technical virtuoso and master of arpeggios

I like how it says guitarists instead of being singular, because there are so many different styles and there isn't one true way to play guitar.

Hm... I don't know about 'best', but some of my favorites:

Kirk Hammett
Tom Morello (Only one mention so far?  * surprise * )
John Petrucci (Dream Theater)
Frank Klepacki (Composer of a ton of C&C music, and also does guitar/drums in some of the songs) and his freaking epic guitar (google it)
Jerry Cantrell (Alice in Chains. Not the best guitarist in the world, but still awesome)
Dino Cazares or Fear Factory (Machine gun guitar riffs FTW)

No, Morello has been mentioned a couple of times, including the very first post by me. Does deserve more credit for his unique sound from RATM, to his solo Nightwatchman folk work.

From start to finish, brother. One of the most underrated. I never really dug Audioslave so much, even though I'm a big fan of Cornell (early) and of course Tom, Brad, and Tim. It's just whenever I heard them play, I would expect Zacks voice to accompany. Just didn't mesh well with me. Kudos to the riff from "Show Me How To Live" as well.

King, Clapton, Young and Satriani are my favs. I dont listen to as much Clapton as I should, but his fingers are magic. Satriani plays like a kid during his first visit to a candy/porn shoppe. King and Young, well, If I need to go there you obviously hate the guitar.

Angus Young would be my favorite hard-rock guitarist though.
